Does anyone know how valuable this is if your undergrad record is not perfect?

I am currently taking real analysis and am also applying to programs. I feel like I may end up with a less than stellar grade in this course. I was just wondering if people know whether programs require you to submit updated transcripts with fall semester grades or specifically ask for grades in certain fall semester courses, in my case real analysis, if they are mentioned in the application.
